ZY102 – Non-Confidential Investors Review 2011

Aripiprazole Oral Once Weekly

ZY102 is a controlled release formulation of the existing second generation atypical antipsychotic agent Aripiprazole. Zysis has re-formulated the existing standard release product to generate an agent which can be dosed orally once weekly. This new controlled release version has completed Phase I bioequivalence testing, and is currently moving into Phase II PET scan/dose ranging studies.

Aripiprazole Oral Once Weekly offers another option for patients struggling with treatment adherence. At present, the only option as a next step is to move onto painful, once-monthly depot injections. This can actually result in worse treatment adherence as the patient can feel alienated, eventually become lost to the system and stop visiting the physician altogether. A Once Weekly Oral Aripiprazole treatment should improve staff-patient relationships rather than cause a deterioration.

The atypical class of antipsychotic agents remains outside any restrictions imposed on choice of individual product prescription in the USA.

ZY102 is currently available for partnering/out-licensing.

Nocturel

Nocturel is a new controlled-release formulation of Trimipramine.

Trimipramine was one of the last tricyclic antidepressants to be developed with excellent anxiolytic, antidepressant and insomnia properties. It has a favourable side effect profile compared to older tricyclics, however it is not easy to prescribe, having to be titrated carefully to avoid hypotension. The tricyclics are still used extensively today for moderate to severe agitation, depression & anxiety.

Insomnia is one of the six cardinal symptoms of anxiety. Apart from its direct anxiolytic activity, Trimipramine is unique in that it works on both quality and quantity of sleep, improving the amount of REM sleep thus providing patients with refreshing rest and therefore improving their state of mind.

Nocturel is a controlled-release reformulation of trimipramine. It has an improved dosing regimen, smoother deliver of the compound, thus making it easier to titrate and should also result in an improved side effect profile for patients.

Its efficacy profile will be better defined with a clear indication in an anxiety disorder providing physicians with a well-defined patient group. Nocturel is currently available for partnering/out-licensing.
